KNIT method inserts large DNA fragments without double-strand breaks
Scientists have developed a genome-editing technique called KNIT (kilobase-scale nickase-targeting) that allows precise insertion of DNA fragments exceeding 10 kilobases using only a single-nick mechanism, avoiding the risks associated with double-strand breaks. The method works across multiple genomic loci and cell types with high efficiency and minimal off-target effects. The study appeared in Nature on 22 July 2026.
